Cryo-EM : Single Particle Imaging

Description

Over the last decade, interest in cryo-electron microscopy has grown due to groundbreaking developments in instrumentation. This has resulted in the explosive growth in the production of structures using Single Particle Analysis (SPA). Apart from the course to teach users how to operate high-end systems in the Nano-Imaging Core Facility (NICF-IP), we now introduce a new course on how to process SPA data to the highest resolution.
This course
•    will teach users how to use the most commonly used reconstruction packages like Relion and CryoSPARC.
•    will be predominantly hands-on and practical’s laced with a few theoretical lessons given by experts from the field on how to validate your data.

This course will also use the "wrapping" package Scipion in which a user can run multiple reconstruction software and cross check the results in a simple and intuitive way. It will be given in the new education center of Institut Pasteur with access to Pasteur servers and computing power.

Participants will be using the Institut Pasteur high performance computing clusters and servers under the supervision of IP – Paris-Cité University instructors and guest teachers Jose Maria Carazo and Carlos Oscar Sorzano, both SPA reconstruction software developers. The course will offer extensive practice on the basis of image processing using a test set. You will be using Relion and Cryo-SPARC followed by an Introduction to Scipion. Participants will have the opportunity of processing the datasets they have collected in NICF-IP. Processing of datasets obtained in other facilities can be discussed with the organizers.  The participant will go in depth on validation of the outcome and how to make sure the results can be trusted.
