Steady-state behavior of endogenous NK cells in the lymph node: view
Ncr1GFP/+xCD11c-YFP mice were adoptively transferred with labeled CD8 T cells. NK cells are pseudocolored in red, CD8 T cells in blue, and DCs in green.

   

Intravital two-photon imaging of Leishmania major-specific CD4 T cells patrolling theinfected dermis: view
Lys-EGFP mice were infected with DsRed-expressing L. major. After one week, mice ere adoptively transferred with parasite-specific T cells. This movie shows an example of a T cell (on the left) that decelerates upon contact with an infected cells. A second T cell (on the right) that do not contact infected cells during the imaging period maintains a high motility. T cells (brown), L. major (red), Lys-GFP (green)


   

Intravital two photon imaging on tumor bearing mice after CTL adoptive transfer: view
Mice bearing established EG7-mYFP tumors and adoptively transferred with activated GFP-expressing OT-I CD8 T cells were subjected to intravital two photon imaging. Time-lapse video revealed that CTLs located within regions in which tumor cells had been eliminated (delineated by a blue dashed line) were very motile. Other CTLs were sequestred at the border of these regions, being engaged in long-lasting interactions with tumor cells. This movie illustrates that intratumoral CTL dissemination occurs concomitantly with tumor cell elimination.


   

Intravital two photon imaging of tumor cell apoptosis during adoptive T cell therapy: view
Mice bearing established EG7-DEVD (a sensor of caspase 3 activity) tumors and adoptively transferred with activated GFP-expressing OT-I CD8 T cells were subjected to intravital imaging. Time-lapse video showed that apoptotic tumor cells (green) were closely associated with CTLs (red) for prolonged period of time. The vast majority of tumor cells that were not contacting CTLs were alive (yellow).

   

Tumor cells undergo apoptosis during their interactions with CTLs: view
Mice bearing established EG7-DEVD tumors and adoptively transferred with activated GFP-expressing OT-I CD8 T cells were subjected to intravital two photon imaging. Time lapse video showing 3 examples of tumor cells undergoing apoptosis while being engaged by antitumor CTLs (red). Note that the color of these tumor cells shifts from yellow to green indicative of increasing caspase 3 activity.

CD4+ T Cells establish long-lived interaction with the first Ag-bearing DC encountered: view
MHC class II-deficient recipients were injected with DCs isolated from female Ubi-GFP mice (green) and were adoptively transferred with SNARF-labeled Marilyn CD4+ T cells (red). After 24hr, mice were prepared for intravital imaging of the popliteal LN. Time-lapse images were acquired with two-photon microscopy. After 11min of imaging, 50μg of Dby peptide was injected intravenously through the jugular vein. Imaging was then continued for 1hr.

   

Zoomed-in view of CD4+ T Cell-DC interactions early after peptide injection: view
