In this paper we show that that the relaxation signal of the water molecule protons, induced by the presence of the paramagnetic manganese ion, which had until then been assigned to the local movements of the molecule complexing the ion, is in fact due to the electronic spin of manganese, at least at room temperature. This observation stopped all use of the manganese ion as a tool to explore the local dynamics of macromolecules, as it showed that the interpretation of all previous research in the domain were wrong

A theory of the epigenesis of neuronal networks by selective stabilization of synapses
Proc Natl Acad Sci U S A (1973) 70: 2974-2978

Abstract: A formalism is introduced to represent the connective organization of an evolving neuronal network and the effects of environment on this organization by stabilization or degeneration of labile synapses associated with functioning. Learning, or the acquisition of an associative property, is related to a characteristic variability of the connective organization: the interaction of the environment with the genetic program is printed as a particular pattern of such organization through neuronal functioning. An application of the theory to the development of the neuromuscular junction is proposed and the basic selective aspect of learning emphasized.

Affinity labeling of the adenosine 5'-monophosphate binding site of rabbit muscle glycogen phosphorylase b with an adenosine 5'-monophosphate-cobalt(3) complex
J Biol Chem (1973) 248: 3241-3247
pdf_file

Abstract: A new affinity labeling method is presented which may be used for enzyme-catalyzing reactions in the presence of magnesium. This method uses the substrate-cobalt(III) analog of the substrate-magnesium complex as a covalent label of the active site. The specific case of rabbit muscle glycogen phosphorylase b, which has been covalently labeled with a AMP-Co(III) derivative at the allosteric site, is discussed. The stoichiometry and the extinction coefficient of the label have been determined. The association of this cobaltic complex to the enzyme is stable. The label destroys the activity on the subunit where it is bound; it slightly affects the binding of orthophosphate and it changes the response to the glycogen substrate from a noncooperative pattern to a cooperative binding. This shows that the label has an effect on the quaternary structure of phosphorylase b. In addition to the covalent binding of the label to the allosteric site, it is shown that its association can be reversed at will, i.e. it can be removed by the addition of small molecules containing thiol groups.

Biochemistry (1976) 15: 1357-1362
pdf_file

Abstract: Formation of the 30S-tRNA initiation complex of Escherichia coli with nonformylated initiator tRNA is stimulated by all three initiation factors and is messenger dependent, whereas the complex formation involving the 70S ribosomes is strongly inhibited by initiation factors when the nonformylated species is used. When the 30S-Met-tRNAfMet complex is first formed and the 50S ribosomal subunit added subsequently, there is no significant inhibition by initiation factors and the nonformylated initiator tRNA is puromycin reactive. This leads to the conclusion that the formylation of the methionyl initator tRNA is only obligatory when polypeptide synthesis is initiated by nondissociated 70S ribosomes.

Toward an understanding of the formylation of initiator tRNA methionine in prokaryotic protein synthesis. II. A two-state model for the 70S ribosome
Biochemistry (1976) 15: 1362-1369
pdf_file

Abstract: The 70S ribosomes can select the proper initiator tRNA between Met-tRNAfMet and fMet-tRNAfMet. Experiments on binding and on formation of aminoacylpuromycin, as a function of magnesium, potassium, or initiation factors, suggest a two-state equilibrium for 70S particles, involving a minor, active conformation and a major one which is not readily active. The formyl group would act as a specific trigger to select the active conformation. Experimental results are interpreted following this simple model and equilibrium parameters, together with kinetic constants of the peptidyltransferase activity, are presented.

This article shows for the first time that the "serine effect" independently discovered by Alföldi is not reversed by threonine or 2-ketobutyrate, but shows a particular sensitivity of bacteria towards serine, for a reason that we just understood (january 6th, 2006)
