  abstract

 

We use X-ray crystallography to study the proteins of protein complexes involved in the different steps of the viral cycle, using several viral model systems. We have provided mechanistic insights into the process of entry of enveloped viruses, introducing and characterising class II viral membrane fusion proteins, with the crystal structures of the flavivirus and the alphavirus envelope proteins in both their pre- and post- membrane fusion conformation. Other results concern viral replication, with the 3D structure of the hepatitis C virus RNA polymerase, and the assembly of viral particles like those of rotaviruses and birnaviruses (which are icosahedra double-stranded RNA viruses) and the HIV-1 capsid protein in complex with an inhibitor of assembly.
