Figure from Pizarro-Cerda et al. Cell Biology Handbook, 2006 Differential immuno-labelling of intracellular versus extracellular bacteria. LoVo cells were infected with Listeria monocytogenes as described in the procedures and extracellular bacteria were distinguished from intracellular micro-organisms by differential immuno-labelling. (A) Extracellular bacteria were labelled with an anti-Listeria serum without permeabilising the LoVo cells. (B) The total population of bacteria (both extracellular and intracellular) were labelled with the same anti-Listeria serum after permeabilising the LoVo cells. Intracellular bacteria are identified as those bacteria that are labelled only after the permeabilisation treatment. (C) Phase image of LoVo cells and Listeria (bar: 5 µm). Arrow head: Intracellular bacteria. Arrows: Extracellular bacteria. |
