Institut Pasteur

This course will be taught in French. However, the protocols book will be written in English
and some presentations will be in English.

This course is devoted to the exploration of the various roles that RNAs molecules are able to play into the gene expression and to the analysis of their regulations. Several lecture will be focused on the multiple roles of RNAs.
The objectives of the practical course are to study some steps of mRNA regulation such as export from the nucleus or stability. To this aim, the quantification of cytoplasmic and nuclear mRNAs in different mutant conditions will be performed at a genomic scale.
Practical course will consist of:
1) purification of ribonucleoproteic complexes;
2) extraction of associated RNAs;
3) hybridization onto micro arrays;
4) analysis of data after scanning;
5)validation of the results by classical approaches such as qRT-PCR.
